Preparing a blood sample Oxygenscan

Use the dedicated Lorrca reagent Oxy ISO (see Appendix - Parts list)

Sample preparation:

  1. Measure RBC level on a Cell Counter (RBC in 106/µL or RBC in 1012/L).
  2. Oxygenscan whole blood volume calculation: 200/RBC = xx µL blood in 5 mL Oxy ISO, this should result in a value between 30 and 200 µL depending on the cell count (level of anemia)
  3. Take the sample from the fridge, bring the sample to room temperature
  4. Gently mix the sample by reversing the primary tube 15 times. Verify that there is no sediment sticking to the bottom of the primary tube. Otherwise, continue mixing until this is dissolved.
  5. Gently pipet the calculated standardized sample volume into the Oxy ISO.
  6. Return the sample to the fridge.
  7. Gently mix the Oxy ISO vial by reversing 40x, use the sample/Oxy ISO solution within 15 minutes. (Oxy ISO is not a storage medium)
  8. Slowly draw 1,6 mL of the sample/Oxy ISO solution in a new(dry) 3 mL syringe. Remove air bubbles.

A blood sample can be kept at room temperature during a limited time. Sickle cell samples can be more fragile than expected.
